Joidenkin masennuslääkkeiden pitkäaikainen käyttö voi aiheuttaa 5-HT<sub>1A</sub> -reseptorin turtumista, joka johtaa negatiiviseen kierteeseen ja lääkehoidon tehon heikkenemiseen. Vortioksetiini toimii 5-HT<sub>1A</sub> -reseptorin agonistina, mikä saattaa nopeuttaa antidepressiivisia vaikutuksia.
