Henderson vaikutti nuorena Detroitin 1950-luvun jazzpiireissä. Kahden vuoden armeijassa vietetyn ajan jälkeen (1960-1962) hän muutti New Yorkiin, jossa häntä opasti musiikissa Kenny Dorham. Henderson liittyi Horace Silverin yhtyeeseen. Lähdettyään yhtyeestä vuonna 1966, hän jatkoi työskentelyä freelancena ja johti myös Kenny Dorhamin ohella big bandiä. Hän soitti useissa Blue Noten 1960-luvun levytyksissä. Hän kuoli vuonna 2001 kärsittyään pitkällisestä keuhkolaajentumasta.
